We present results of our recent investigations on the chemical conditions for the formation of mixed self-assembled monolayers of thiol-modified oligonucleotides and mercaptohexanol deposited onto gold surfaces. Using high-resolution X-ray photoemission spectroscopy we found that the spatial density of DNA strands can be varied in a controlled manner within a wide range by the concentration of MgCl2 in the immobilization buffer. We were also able to illuminate the question how the DNA probes affect the quality of the mercaptohexanol SAM more deeply. In particular, the use of aqueous instead of ethanolic mercaptohexanol solutions with a more moderate mercaptohexanol concentration is demonstrated to be favorable for the formation of high-quality mixed monolayers.
