Fluorescence correlation spectroscopy is a method to study the self-diffusion of fluorescence-labeled molecules. With this method, the detection volume is very small (ca. 1 µm3) and the measurements are carried out on the basis of a single molecule in the detection volume.

We have studied aqueous solutions of water-soluble poly(methyloxazolin) (PMO) labeled with aminofluorescein as a function of concentration. In this way, the influence of polymer concentration on the diffusivity is studied on a microscopic scale. These results are compared to the diffusion of fluorescence-labeled latices in PMO solutions.
