The activity of a single helicase opening a double strand DNA can be monitored in real time using a magnetic trap technique to pull on a single DNA molecule and observe its increase in extension as it is unwound into two ssDNAs. This observation allows one to measure the rate of unwinding, the enzyme’s processivity, on-time and step size as a function of force and ATP concentration. These single molecule measurements differ markedly from bulk measurements of the activity of the helicase we study. We shall explain why.
