0pt nohyphenation Circular scanning Fluorescence Correlation Spectroscopy (csFCS) is a calibration-free alternative to conventional static point FCS [1,2]. Not only does csFCS allow for the direct determination of the diffusion coefficient, but, in case of quasi-planar membranes, also avoids potential errors originating from imprecise axial positioning. Previously, csFCS has been used to measure diffusion in giant unilamellar vesicles (GUVs), serving as a model of free-standing lipid membranes [3]. Despite the immanent importance of model membranes, their temperature-dependent lipid mobility has been subject to only few studies. Here, we report on our progress of employing csFCS to systematically characterize the diffusion in GUVs over a wide range of temperatures.
