Recently, the layer-by-layer technique provided a convenient way to form functional and organized thin films in predetermined ways. Here, this method was applied for the formation of multilayer films of DNA and polypropyleneimine dotriacontaamine dendrimer, generation 4.0 (G4), which are discussed as carriers for nonviral gene delivery systems. An exponential growth of the multilayer film thickness with increasing layer number was found. The internal structure of the multilayers, which was investigated by X-ray reflectivity and grazing incidence diffraction, shows the same liquid crystalline structure as it was found recently in bulk complexes. The morphology of the multilayers and the surface roughness were further studied by atomic force microscopy. The exponential growth can be attributed to the combination of the factors of increasing surface roughness during the adsorption of DNA layers, the molecular diffusion during the adsorption of G4 layers, and the complexation during the buildup process.
